Clarinex (desloratadine) is a leading prescription antihistamine specifically formulated to deliver potent, non-sedating relief from the most stubborn allergy symptoms and chronic idiopathic urticaria. As a selective peripheral H1-receptor antagonist, it represents a refined advancement in antihistamine therapy, offering targeted action with a well-established clinical profile. Trusted by allergists and healthcare providers globally, Clarinex works by effectively blocking histamine, a key substance your body produces during an allergic reaction, providing sustained 24-hour relief from a single daily dose. This allows patients to regain control over their daily lives without the disruptive drowsiness associated with older generation allergy medications.
Features
- Active Ingredient: Desloratadine (5 mg per tablet).
- Pharmacological Class: Second-generation, selective peripheral H1-receptor antagonist.
- Duration of Action: Provides 24-hour symptom relief from a single dose.
- Formulations: Available in film-coated tablets, orally disintegrating tablets (RediTabs), and a syrup formulation.
- Onset of Action: Symptom relief begins within one to three hours post-administration.
- Bioavailability: Not significantly affected by food intake, allowing for flexible dosing.
Benefits
- Sustained 24-Hour Relief: Effectively controls a wide range of allergy symptoms—including sneezing, runny nose, itchy/watery eyes, and itchy palate—for a full day with a single dose, supporting consistent compliance and symptom management.
- Minimal Impact on Central Nervous System: Its high selectivity for peripheral H1-receptors significantly reduces its ability to cross the blood-brain barrier, drastically lowering the incidence of sedation and cognitive impairment compared to first-generation antihistamines.
- Improved Quality of Life: By providing reliable control over debilitating allergy symptoms and the distressing welts and itching of chronic hives, it enables patients to maintain normal sleep patterns, work productivity, and social activities.
- Proven Efficacy in Multiple Conditions: Demonstrated effectiveness in treating both seasonal allergic rhinitis (hay fever), perennial allergic rhinitis (year-round allergies), and chronic idiopathic urticaria, making it a versatile therapeutic option.
- Favorable Safety and Tolerability Profile: Supported by extensive clinical trial data, showing a low incidence of adverse events, which are typically mild and transient in nature.
Common use
Clarinex is indicated for the relief of the symptoms associated with:
- Seasonal Allergic Rhinitis: Symptoms including sneezing, rhinorrhea (runny nose), nasal congestion, pruritus (itching) of the nose/palate/throat, and itchy, watery/red eyes in patients 2 years of age and older.
- Perennial Allergic Rhinitis: Year-round allergy symptoms caused by indoor allergens such as dust mites, pet dander, or mold, in patients 6 months of age and older.
- Chronic Idiopathic Urticaria: For the treatment of unremitting hives (wheals) and associated pruritus (itching) of unknown cause, in patients 6 months of age and older. It effectively reduces the size, number, and duration of hives and alleviates itching.
Dosage and direction
Dosage is based on the patient’s age and medical condition. The tablet should be taken orally, with or without food, typically once a day.
- Adults and Adolescents (12 years and older): 5 mg (one tablet) once daily.
- Children (6 to 11 years): 2.5 mg (one-half of a 5 mg tablet) once daily. The oral syrup formulation (0.5 mg/mL) is often preferred for accurate dosing in this age group (5 mL or 1 teaspoonful).
- Children (12 months to 5 years): 1.25 mg (one-quarter of a 5 mg tablet or 2.5 mL of syrup) once daily.
- Children (6 to 11 months): 2 mL of syrup once daily.
- Clarinex RediTabs (orally disintegrating tablets) should be placed on the tongue and swallowed with or without water. The tablet disintegrates within seconds.
Precautions
- Prior to starting Clarinex, inform your healthcare provider of your complete medical history, especially of kidney or liver disease. Dose adjustment may be necessary for patients with significantly impaired renal or hepatic function.
- While Clarinex is classified as a non-sedating antihistamine, individual responses can vary. Exercise caution when driving, operating machinery, or engaging in activities requiring alertness until you are sure the medication does not cause drowsiness.
- The safety and effectiveness of Clarinex in children under 6 months of age have not been established.
- Use during pregnancy only if clearly needed. Desloratadine is excreted in breast milk; therefore, caution is advised if Clarinex is administered to a nursing woman. A decision should be made whether to discontinue nursing or discontinue the drug.
Contraindications
Clarinex is contraindicated in patients with a known hypersensitivity to desloratadine, loratadine, or any of the inactive ingredients in the formulation.
Possible side effect
In clinical trials, side effects were generally comparable to placebo. The most commonly reported side effects (occurring in ≥2% of patients and greater than placebo) in adults and adolescents were:
- Fatigue
- Dry mouth
- Headache
- Somnolence (drowsiness)
- Pharyngitis
- Myalgia (muscle pain)
- Dysmenorrhea (menstrual pain)
- Nausea
- Dizziness In children, the most common side effects included diarrhea, fever, and insomnia. This is not a complete list of possible side effects. Patients should consult their physician for medical advice about side effects.
Drug interaction
No clinically relevant drug interactions have been found in controlled studies. Specifically:
- Clarinex has been co-administered with azithromycin, ketoconazole, erythromycin, and fluoxetine without evidence of significant interactions or increased adverse effects.
- No meaningful interaction was observed with alcohol; however, as a general precaution, alcohol should be avoided as it may increase the risk of drowsiness.
- Always inform your doctor of all prescription, non-prescription, and herbal products you are using before starting any new medication.
Missed dose
If you miss a dose of Clarinex, take it as soon as you remember. If it is almost time for your next dose, skip the missed dose and resume your usual dosing schedule. Do not double the dose to catch up.
Overdose
In the event of a suspected overdose, symptomatic and supportive treatment is recommended. In clinical trials, doses up to 45 mg (9 times the recommended daily dose) were administered and resulted in an increased incidence of headache, somnolence, and tachycardia (fast heart rate). Inducing vomiting is not recommended. In case of overdose, contact a poison control center or emergency room immediately.
Storage
- Store Clarinex tablets and RediTabs at room temperature (20°-25°C or 68°-77°F) in a tightly closed container.

Reviews
“After struggling with year-round allergies that made me feel constantly fatigued and congested, my allergist prescribed Clarinex. The difference was night and day. The 24-hour relief is consistent, and I experienced zero drowsiness, which was a major problem for me with over-the-counter options. It has genuinely given me my quality of life back.” – Sarah K., Verified Patient
“As a clinician, I find Clarinex to be an exceptionally reliable second-line antihistamine for patients who do not achieve full symptom control with loratadine or cetirizine. Its metabolic profile and lack of significant drug interactions make it a safe choice for many of my patients, including those on multiple medications. The clinical data supporting its efficacy in both allergic rhinitis and CIU is robust.” – Dr. Evan R., Allergist-Immunologist
“My son has severe chronic hives, and the itching was unbearable for him. Since starting on Clarinex syrup, the outbreaks have become less frequent and far less intense. He’s finally sleeping through the night and is much happier during the day. We are very grateful for this medication.” – Mark T., Parent
